NTFS is a more complex file system, it's main benefits include security, compression and stability. You won't notice much of a performance increase, if any, due to the increased overhead.
NTFS should work with any of your exisiting programs unless you have any which bypass the WinAPI and access the disk directly (very unlikely).
You can convert using the following command from a command window: "convert c: /FS:NTFS"

I used that command from a command window Lucifer, but it then asks for a volume label, how do I find my volume label?
Thanks

Type "convert /?" at the command prompt to get the correct syntax -- the command I gave was for Windows 2000 and it might be slightly different for XP.
